Sharkstooth Combs: Sharkstooth combs
provide for "side-by-side" sample application, making
sequencing result analysis easier by eliminating the space between
sample wells. This method requires the formation of a single well,
into which the points of the sharkstooth comb are inserted. A
single well can be formed by either using a 1-well casting comb
or the back straight edge of the sharkstooth comb. Sharkstooth
combs for the 20cm wide unit are available in three thicknesses
(0.2mm, 0.4mm, and 0.5mm) with 24 wells (5.92mm point to point),
34 wells (4.5mm point to point), 38 wells (4.02mm point to point),
52 wells (3.0mm point to point), and 70 wells (2.25mm point to
point). Sharkstooth combs for the 33cm wide unit are available
in three thicknesses (0.2mm, 0.4mm, and 0.5mm) with 46 wells (5.92mm
point to point), 62 wells (4.5mm point to point), 68 wells (4.02mm
point to point), 94 wells (3.0mm point to point), and 126 wells
(2.25mm point to point).
Rectangular Tooth Combs: Rectangular tooth combs are precision-machined polycarbonate and are available in a wide variety of sizes for both 20cm and 33cm wide units. Sequencing combs for the 20cm wide unit are available in three thicknesses: 0.2mm, 0.4mm, and 0.5mm; and are offered with 8, 12, 14, 16, 17, 20, 28, 32, or 35 teeth. Teflon¨* combs for protein or preparative gels can also be ordered for the 20cm unit with thicknesses of 0.75mm, 1.0mm, 1.5mm, 2.0mm, or 3.0mm with 1, 2, 3, 4, 8,12, 14, 16, 17, 20, 28, 32, or 35 teeth. Sequencing combs for the 33cm wide sequencer are available in three thicknesses: 0.2mm, 0.4mm, or 0.5mm, and are offered with 8, 16, 24, 31, 34, 50, 60, or 63 teeth. Teflon¨ combs for preparative or protein gels for the 33cm wide unit can be ordered with thicknesses of 0.75mm, 1.0mm, 1.5mm, 2.0mm, or 3.0mm with 3, 5, 6, 8, 12, 16, 24, 31, 34, 50, 60, or 63 teeth.
Sooner Scientific offers
the researcher the option to have numbers printed above the wells
of the sharkstooth combs. This convenient feature allows the researcher
to keep track of which sample goes into each application site
during loading. Shown above with microtrough glass plates. These
specially designed plates enable quick and convenient loading
of small amounts of sequencing sample.
Combs for Multichannel Pipettor Loading: Sooner Scientific has designed rectangular tooth and sharkstooth combs with microtitre plate spacing for multi-channel pipettor loading. Rectangular combs with this feature are the 17 and 35-well combs for the 20cm wide units and the 31 and 63-well combs for the 33cm wide units. The Sharkstooth combs that can be loaded with a multichannel pipettor are the 2.25mm, 3.0mm, and 4.5mm point to point combs. The 2.25mm point to point combs correspond to our 126-well comb for the 33 cm wide sequencers or our 70-well comb for the 20cm wide sequencer, and a multichannel pipettor would load every 4th well. The 3.0mm point to point combs correspond to our 94-well comb for the 33 cm wide sequencers or our 52-well comb for the 20cm wide sequencer, and a multichannel pipettor would load every 3rd well. The 4.5mm point to point combs correspond to our 62-well comb for the 33 cm wide sequencers or our 34-well comb for the 20cm wide sequencer, and a multichannel pipettor would load every other well.
Spacer Sets -
Linear Wedge Spacers: Linear wedge spacers
are offered in 3 different ratios, 1:2, 1:3, and 1:4. They are
available to fit 42cm, 52cm, and 62cm glass plate lengths for
Sooner Scientific's sequencing equipment and can be special ordered
to fit other suppliers' equipment as well. These linear wedge
spacers are precision machined from a single piece of grey PVC.
A bottom spacer for the wedge sets is available as an option which
some researchers have found useful for preventing leakage. The
thinner top part of the spacers can start at either 0.2mm (.010")
or 0.4mm (.015") for all three different ratios of wedge
spacers.
Flat Spacer Sets: Spacer sets for both the 20cm wide and the 33cm wide sequencers are available in three thicknesses (0.2mm, 0.4mm, and 0.5mm) and for gel lengths of 22cm, 32cm, 42cm, 52cm, 62cm, 82cm, and 102cm. Spacer sets for preparative or protein gels can be ordered with thick-nesses of 0.75mm, 1.0mm, 1.5mm, 2.0mm, and 3.0mm.
Exponential Wedge
Spacers: Exponential wedge spacers differ from linear in that
two sets of spacers are used for each gel. The first is a flat
spacer set, followed by a wedged pair. The spacers are assembled
end to end on the glass plate and the gel is poured using standard
techniques. This system maximizes the "wedge spacer effect",
allowing routine sequence penetration of 400-800 bases. The exponential
wedge spacer sets are available in two different ratios, 1:3 and
1:5. They are available to fit 42cm, 52cm, 62cm, 82cm, or the
102cm glass plate lengths for Sooner Scientific's sequencing equipment.
Glass Plates and Aluminum Heat Dispersion Plates - Glass Plates: Glass plates sets are 3/16" thick and can be ordered in lengths of 22cm, 32cm, 42cm, 52cm, 62cm, 82cm, or 102cm. Standard notched plate sets are provided with the kits. Unnotched plate sets with detached rubber ears can also be ordered. German tempex glass (pyrex) is available for special applications. All glass plates are edge sanded and polished for safety and durability.
Aluminum Heat Dispersion Plates: Ultra-flat machine-surfaced 1/4" thick aluminum plates are safety coated and are electrically non-conductive. They can be ordered to clamp behind the glass plate sandwich during electrophoresis. The use of these heat dispersion plates effectively eliminates the "smile effect" by evenly distributing the heat generated during electrophoresis. They also keep pre-run times to a minimum, unlike those systems which use extended upper buffer chambers. Aluminum plates are available for all glass plate sizes for the adjustable systems. (Dedicated Height and Dual Dedicated Sequencers include built-in aluminum heat dispersion plates.)
White Clamps -
Sooner Scientific now has available molded
polypropylene spring clamps for use with the Dedicated Height
and Adjustable Nucleic Acid Sequencers. These clamps are designed
specifically for this application and feature a stainless steel
spring. There are two different types of clamps for sequencing
applications. The clamps shown above are Cat. # FOB-0002 and are
used for holding thinner sandwiches together. Cat. # FOB-0001
is for clamping the glass plates to the upper reservoir and holding
the aluminum heat dispersion plates in place.
Gel Sealing Tape - Yellow mylar tape
is available for the sealing of sequencing glass plate sets. Each
roll of tape is 72 yards long with choices of 1" width (Cat.
# FS-72-10) or 1.5" width (Cat. # FS-72-15).
Quick-Disconnect Coupling -
The quick-disconnect tubing coupling (body,
Cat. # PCB-B and insert, Cat. # PCB-I) is a convenient accessory
for simplifying buffer disposal. This coupling features an automatic
shut-off valve and can be ordered to replace the standard tubing
adapters supplied with Sooner Scientific Sequencing Systems. The
coupling insert with shut-off valve threads into the buffer reservoirs,
while the coupling body attaches to the drainage tubing. To drain
reservoir, simply snap coupling body onto coupling insert and
the valve will open. When the coupling is disconnected the valve
automatically closes.
Bar Clamp -
Sooner Scientific features bar clamps for use
with the Adjustable Sequencing System. (Cat. # FOB-1000 for the
20cm wide units, and Cat. # FOB-2000 for the 33cm wide units.)
Bar clamps are standard with the Dedicated Height and Dual Dedicated
Height Sequencing Systems. The clear acrylic bar clamp evenly
compresses the glass plate sandwich to the upper reservoir gasket,
replacing bulldog spring clamps. The bar clamp is particularly
useful for two reasons:
