SeaQualite SQA-10025, SQA-11025, SQA-12100, SQA-13025, AND SQA-14025 are low melting agaroses derivatized by organic synthesis that offer the user lower temperatures and important reductions in both gelling and melting points. Essentially, the process generates methoxylates from the basic agarose structure.
Low melting agaroses are very useful in operations in the laboratory that involve cell and the tissue cultures from animal or plant sources. The lower gelling and melting temperatures enable the correct digestion of DNA and RNA in the melting gel, with efficient recovery of the particles without running the risk of denaturization.
ADVANTAGES
1. Highest gel strength in the market, 80% superior to existing products. This substantially improves reliability in handling; the gel does not break easily.
2. Excellent digestibility of DNA and RNA because of the absence of inhibitors.
3. Post-extraction purification of DNA and RNA is not a requirement.
4. Better separation and increased recovery rates of DNA and RNA fragments.
LOW MELTING AND LOW GELLING POINT AGAROSES GENERAL CHARACTERISTICS OF LOW MELT TYPES:
- Higher clarity (gel transparency) than standard melting and gelling temperature agaroses.
- Lower melting and gelling temperatures.
- And, like all low melting and gelling point agaroses in the market, they have a lower gel strength than standard melting agaroses.
SeaQualite SQA-10025, SQA-11025, SQA-12100, SQA-13025, AND SQA-14025: These agaroses have a greater sieving properties than standard melting agaroses and are recommended for separation of DNA/RNA fragments greater than 1000bp as well as proteins in preparative electrophoresis.
-The low melting temperature(<65 °C) allows for recovery, without damage, of nucleic acids below there denaturing temperatures.
-The low gelling temperature(24 °C-28 °C) assures the agarose will be in a liquid state at a temperature range where in-gel manipulations (such as enzyme digestion, ligation, DNA labeling, nick translation, random priming, PCR, etc.), can be performed without prior purification of the DNA from the gel slice.
-Tissue culture and viral plaque applications.
-No detectable DNase or RNase activity.
-May be used with all buffer systems at normal concentrations (1%).

SeaQualite 11025 agarose is a low melting temperature agarose with the highest resolving capacity for large DNA fragments, >1000bp, including PCR products.This agarose is genetic quality tested and certified. This ensures the in-gel applications can be performed in remelted agarose, avoiding difficult DNA extraction steps. SeaQualite SQA-11025 agarose is ideal for digestion by agarose enzymes, which makes it very easy to recover large DNA fragments suitable for cloning or enzymatic processing.
Applications:
-Electrophoresis of DNA fragments >1000bp.
-In-gel enzymatic processing (digestion, ligation, PCR.).
-Preparative electrophoresis.
-Analysis and recovery of large DNA fragments for further applications.
Functional Test:
-DNA resolution: bands appear sharp and finely resolved.

SeaQualite SQA-12100 agarose is a low melting temperature agarose with the highest resolving capacity for DNA fragments smaller than 1000 bp, especially PCR products ranging from 200 to 800 bp. This agarose is genetic quality tested and cedrtified. This ensures that in-gel applications can be performed in remelted agarose, avoiding difficult DNA extraction steps. SeaQualite SQA-12100 agarose is ideal for digestion by agarose enzymes, making it very easy to recover small DNA fragments suitable for cloning or enzymatic processing. SeaQualite SQA-12100 agarose can be used at high concentrations, forming gels with excellent clarity and a higher sieving capacity than standard melting agaroses. Due to their high gel strength, SeaQualite SQA-12100 agarose gels are very easy to handle, even at concentrations as low as 2%.

-The difference between SeaQualite SQA-10025, SQA-11025, SQA-12100, and SeaQualite SQA-13025, AND SQA-14025 lies in the fact that SeaQualite SQA-13025, AND SQA-14025 has a more open structure, which allows for a slightly faster migration.
-Due to its low gelling point (24°-28 °C) it is recommended to place the gel for one hour at 4°C before use to assure maximum resolution.
SeaQualite SQA-13025 LOW MELT
This agarose has their gelling points (<17°C) and gel strengths lower than the standard melt types. They have to be formulated at greater than 1.2% gel concentrations because at lower concentrations, the gels of these agaroses are very fragile and cannot be handled properly.
At the same gel concentrations, SQA-13025 agarose has more sieving capacity than the low melt types SQA-10025, SQA-11025 and SQA-12100.
-Recommended for Capillary Electrophoresis, hybridoma and tissue cells cloning, preparation of plant protoplasts, viral plaque and anchorage independent growth assays.
- No detectable DNase and RNase activity.
-May be used with all buffer systems.
As the gelling temperature of this agarose is lower than room temperature, it is necessary that gelling be carried out at 4°C overnight.
SeaQualite SQA-14025
This unique agarose has the lowest gelling point (< 13°C) currently available in the world and are recommended for tissue and cell culture.
Especially recommended for Capillary Electrophoresis.
